Head and neck cancer (HNC) kills half a million people every year, with 1.5 million new cases diagnosed worldwide. Following initial treatment recurrence rate is high ~40–70% and current treatment options for recurrent HNC (rHNC) show poor efficacy and severe side effects , with a median survival of only 10 months. We have developed the first biologically targeted radiotherapy, called BNCT/TB0010, which uses tumour-targeting antibodies to deliver boron (10B) specifically to EGFR positive tumour cells for effective local control while sparing normal tissues. . TB0010, a second-generation boron 10B carrier shows revolutionary properties to overcome current issues with first-generation 10B carriers especially targeted accumulation in cancer cells with minimal side effects and low invasiveness, representing a potentially lifesaving option for patients with high unmet need in rHNC. BIOTARGET0010 aims to advance the clinical development of biological tumour-targeting radiotherapy with a multicentre Phase 1b/2 trial in EGFR positive rHNC patients, demonstrating safety and preliminary efficacy of TB0010/BNCT as a basis for pivotal trials. Our cohesive interdisciplinary team with relevant experience and expertise, and appropriate infrastructure ensures the successful delivery of BIOTARGET0010. Our consortium covers expertise in nuclear physics, chemistry and biology, together with , clinical oncology and regulatory affairs, drug manufacturing, HNC, communication and patients’ engagement.
