IDPexec will transform how the community studies intrinsically disordered proteins (IDPs). IDPexec gathers leading groups in Europe, North America and Japan to deliver a standards-first, end-to-end workflow that turns heterogeneous evidence into trustworthy, functionally interpretable conformational ensembles. WP1 extends and operationalises community standards and ontologies, e.g. MIADE for experimental reporting, IDPO with GO/ECO for semantics. Curated records and ensembles will be deposited and cross-linked across databases (e.g. DisProt, PED, UniProt) to ensure FAIR access. WP2 scales high-quality AI-assisted curation and adds evolutionary context (e.g. multiple sequence alignments, phylogenies and bulk feature) to prioritise conserved IDPs, identify lineage-specific segments and build gold-standard datasets. WP3 delivers integrative ensemble generation by combining multi-scale simulations with machine-learning guidance and rigorous refinement against NMR, EPR, SAXS/SANS and smFRET using Bayesian/maximum-entropy approaches with explicit uncertainty quantification. Open, containerised software and interoperable workflows will be released to the community. WP4 establishes a benchmarking and validation framework with preregistered tasks, multi-criteria metrics (e.g. fit to observables, uncertainty) and public leaderboards—closing the loop between method development and biological insight (e.g., phase separation, transient secondary structure, mutation sensitivity). International secondments drive bidirectional knowledge transfer among curators, experimentalists and modellers. Training events build a skilled workforce fluent in standards, FAIR data stewardship and integrative modelling. By delivering calibrated forward models and uncertainty-aware refinement, and institutionalising open benchmarks, IDPexec will raise the bar for IDP research, accelerate adoption in academia and industry, and position Europe as a global hub for IDP data, methods and training.
